MA3-933 detects the junctional sarcoplasmic reticulum (SR) protein from rabbit and guinea pig; moderately in canine and rat; and weakly in mouse.
MA3-933 has been successfully used in Western blot and immunofluorescence procedures. By Western blot, this antibody detects an ~90 kDa protein representing junctional SR in rabbit skeletal muscle extracts. Immunofluorescence staining of junctional sarcoplasmic reticulum with MA3-933 results in an increase in intensity in fast (Type II) skeletal muscle over slow (Type I) skeletal muscle, and does not cross-react with cardiac muscle.
The MA3-933 antigen is rabbit skeletal muscle triads.
The junction between the transverse tubules (T-tubules) and the sarcoplasmic reticulum (SR) of skeletal muscle is called the triad. At the triad, dihydropyridine receptors (DHPR) of the T-tubule serve as voltage sensors in excitation-contraction coupling, while ryanodine receptors (RyR), the calcium release channels, exist in the membrane of the terminal cisternae of the SR. It is thought that during slow phase depolarization of the T-tubule, a third protein, triadin (95 kDa) transmits electrochemical signals to the SR through direct interaction with both DHPR and RyR. Another skeletal muscle specific 90 kDa protein has also been localized to the this junction. This protein is phosphorylated by an intrinsic protein kinase in triads suggesting that it may play an important regulatory or structural role in the skeletal muscle triad junction.
