DAPK1 is a large, calmodulin-dependent kinase involved in gamma-interferon induced programmed cell death and is a potential tumor suppressor. DAPK1 is involved in multiple cellular signaling pathways that trigger cell survival, apoptosis, and authophagy. It regulates both type I apoptotic and type II autophagic cell death signal, depending on the cellular setting. Type I apoptotic cell death is caspase-dependent, type II authophagic cell death is caspase-independent. DAPK1 phosphorylates PIN1 resulting in inhibition of its catalytic activity, nuclear localization, and cellular function. It also phosphorylates TPM1, STX1A, PRKD1, BECN1, TSC2, RPS6, MYL9, and DAPK3.
