Description: This 5E7 monoclonal antibody recognizes mouse B lymphocyte-induced maturation protein-1 (Blimp-1), which is also known as Prdm1. Blimp-1 is a transcriptional repressor with five zinc finger domains. Blimp-1 is expressed by terminally differentiating B and T cells. Blimp-1 is also expressed by some dendritic cells, and some NK cells. This transcriptional repressor is required for B cell differentiation into plasma cells and serves as a critical antagonist of CD4+ follicular helper T cell differentiation. While Blimp-1 is not required for priming and development of T and B cell effector function, it is critical for effector cell terminal differentiation. Blimp-1 promotes effector molecule secretion while suppressing proliferation. Blimp-1 also acts as a transcriptional antagonist of Bcl-6 and inhibits genes that are related to memory cell differentiation.
Applications Reported: The 5E7 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested: The 5E7 antibody has been tested by flow cytometric analysis of splenocytes isolated from sheep red blood cell-immunized mice. This can be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Blimp-1 (B lymphocyte maturation protein-1) is a nuclear protein with five zinc fingers in its carboxy terminus. It is a transcriptional repressor that is both required and sufficient to trigger terminal differentiation of B lymphocytes and monocyte/macrophages. Blimp-1 is expressed in, and may be required for terminally differentiated epithelial cells, retinal neurons and other lineage cells. It has also been implicated in the repression of beta-interferon (IFN-beta) and c-myc gene expression.
