Description: The monoclonal antibody 2G7 reacts to CD84, a transmembrane protein of the Ig SLAM superfamily. Expression of CD84 is found on mature B cells, a subset of T cells (mainly CD4+CD45RA+), monocytes and platelets. CD84 is found on a percentage of human hematopoietic progenitor (CD34+) cells. Signaling through CD84 via homotypic interactions involves phosphorylation and recruitment of SAP and EAT-2 leading to production of IFNgamma and T cell activation.
Applications Reported: This 2G7 antibody has been reported for use in flow cytometric analysis and immunohistology staining of frozen tissue sections.
Applications Tested: This 2G7 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Filtration: 0.2 µm post-manufacturing filtered.
CD84 is expressed on mature B cells and on B cell lines, including pre B cell lines, but not on plasma cell lines. Immunohistochemical studies demonstrated that CD84 strongly expressed on tissues macrophages. CD84 is also highly expressed on platelets and, at low levels on peripheral blood T cells. It is a highly glycosylated protein and belongs to immunoglobulin superfamily. It may play a role in leukocyte activation.
