HAH 002-01-02 has specificity for butyrylcholinesterase from human serum or plasma. Crossreactivity to BtChE from other species has not been tested.
HAH 002-01-02 has been successfully used in ELISA techniques.
HAH 002-01-02 reacts with BtChE from normal serum when tested in sandwich ELISA. This antibody can be used as both the capture and detection antibody or an alternative antibody against BtChE can be used as capture antibody. In addition, it can be used in crossed immunoelectrophoresis followed by incubation of the gel with HAH 002-01-02. In immunoblotting after SDS-PAGE no reaction is seen with either the reduced or with the non-reduced form of BtChE. No reaction is seen with acetylcholinesterase from human nervous tissue and erythrocytes.
The HAH 002-01-02 immunogen is Butyrylcholinesterase found in human plasma cells.
Mutant alleles at the BCHE locus are responsible for suxamethonium sensitivity. Homozygous persons sustain prolonged apnea after administration of the muscle relaxant suxamethonium in connection with surgical anesthesia. The activity of pseudocholinesterase in the serum is low and its substrate behavior is atypical. In the absence of the relaxant, the homozygote is at no known disadvantage.
