Description: The NK92.39.1 monoclonal antibody reacts with human CD96 also known as TACTILE (T cells-activated increased late expression). CD96 is a type I membrane protein of the Immunoglobulin Superfamily. CD96 promotes NK cell adhesion to cells expressing the polio virus receptor (CD155). Therefore CD96 is an additional receptor for CD155. After binding of PVR to CD96, the PVR can be transferred to the target cell and CD96 can be internalized with bound PVR. This internalization may lead to cell death of the NK cell. This phenomenon may be significant in tumor cells which express high levels of PVR. Furthermore expression of CD96 has been found in the majority of CD34(+)CD38(-) Acute Myeloid Leukemic cells.
The NK92.39.1 antibody can block binding of soluble CD155 to NK cell lines.
Applications Reported: This NK92.39 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This NK92.39 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 µL (0.5 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.

CD96 is a cell adhesion receptor also known as TACTILE. A member of the Ig superfamily, CD96 is a single pass transmembrane protein consisting of three extracellular Ig-like domains and a proline/serine/threonine rich region. CD96 is expressed on resident monocytes and T cells at all developmental stages (including gamma-delta T cells, thymocytes, and NK cells). In the mouse, this receptor is not expressed on B cells. Unlike in humans, mouse CD96 does not exist as alternative splice variants. Reports indicate that CD96 binds CD155/poliovirus receptor and Nectin-1 to mediate NK cell adhesion to target cells and cytotoxicity.
