Description: The monoclonal antibody 5E5 recognizes human CD98hc protein also called CD98 heavy chain, FRP-1 (fusion regulatory protein-1 ) or 4F2. CD98 is a disulfide-linked complex composed of a heavy chain (a Type II membrane protein) and a light chain of which there are more than six different chains (LAT1, LAT2, y+LAT1, y+LAT2, Asc-1, xCT). CD98 can associate directly or indirectly with beta 1 integrins, EpCAM and CD147 (EMMPRIN). The function of CD98 is thought to involve amino acid transport, cell fusion concomitantly with increased plasma membrane expression and integrin mediated adhesion. Expression of CD98 is broad including non-hematopoietic cells. In peripheral blood cells, monocytes expression is high while lymphocytes have intermediate levels and granulocytes express very little.
Applications Reported: This 5E5 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This 5E5 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.
CD98 (4F2) is a type II transmembrane glycoprotein which serves as the heavy chain of the heterodimeric amino acid transporters (HATs). CD98, linked to various light chains by disulfide bond, is responsible for cell surface expression and basolateral localization of this transporter complex in polarized epithelial cells and also interacts with beta1 integrins and increases their affinity for ligand. Besides its roles in amino acid transport, CD98 is thus involved in cell fusion and activation. It is implicated in regulation of cellular differentiation, growth and apoptosis.
