Immunofluorescence analysis of CDK1 was performed using 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.
Target Information
CDK1 or Cell Division Control protein 1 is essential for the completion of START, the controlling event in the cell cycle that is required to initiate mitosis. CDK1 is a catalytic subunit of a protein kinase complex, called the M-Phase Promoting Factor that induces entry into mitosis and is universal among eukaryotes. Phosphorylation of Bcl-2 in G2/M phase-arrested cells following photodynamic therapy with hypericin involves a CDK1-mediated signal and delays the onset of apoptosis. Therapeutic potential of CDK inhibitor NU2058 in androgen-independent prostate cancer has also been demonstrated.
