Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human Fractalkine (CX3CL1). Anti-Human Fractalkine (CX3CL1)-specific antibody was purified by affinity chromatography employing an immobilized Human Fractalkine (CX3CL1) matrix.
Sandwich ELISA: To detect Human Fractalkine (CX3CL1) by sandwich ELISA (using 100 µL/well antibody solution) a concentration of 0.5-2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human Fractalkine (CX3CL1) (500-P98Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human Fractalkine (CX3CL1).
Western Blot: To detect hFractalkine by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant hFractalkine is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
Chemokines are a family of proteins associated with the trafficking of leukocytes in immune surveillance and inflammatory cell recruitment. They are classified based on the positions of key cysteine residues. CX3CL1 is a CX3C chemokine known to induce adhesion and migration of leukocytes mediated by a membrane-bound and soluble form respectively. Recent experiments have shown that CX3CL1 can suppress the production of nitrous oxide, interleukin-6, and TNF-a in activated microglia and neuronal cells, suggesting that it may act as an intrinsic inhibitor against neurotoxicity by activated microglia. Its receptor, CX3CR1, also functions as a co-receptor for HIV-1 and HIV-2 envelope fusion and virus infection, which can be inhibited by CX3CL1.
