Knockout of FANCD2 was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, AssayID CRISPR718017_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.
Target Information
FANCD2 (Fanconi anemia group D2 protein) is required for maintenance of chromosomal stability. It promotes accurate and efficient pairing of homologs during meiosis. FANCD2 is involved in the repair of DNA double-stranded breaks, both by homologous recombination and single-stranded annealing. It is required for the targeting/stabilization of BLM to non-centromeric abnormal structures induced by replicative stress. FANCD2 promotes BRCA2/FANCD1 loading onto damaged chromatin. Defects in the FANCD2 gene result in Fanconi anemia complementation group D2. This disorder affects all bone marrow elements resulting in anemia, leukopenia, and thrombopenia.
