Description: This 2A9-mIkaros monoclonal antibody reacts with mouse Ikaros, the 70-kDa founding member of the Ikaros family of C2H2-type zinc finger transcription factors. Ikaros is expressed in hematopoietic progenitor cells, thymocytes, mature T and B cells, monocytes, macrophages, and natural killer cells. This protein has also been shown to be expressed in non-immune cells such as neurons. Studies indicate that Ikaros can interact with other family members, namely Aiolos and Eos. Ikaros is involved in thymocyte development, B cell differentiation, cell cycle control, chromatin remodeling, and tumor suppression. Moreover, Ikaros has been implicated in leukemia and lymphoma. Ikaros has been demonstrated to regulate expression of Tdt, CD8a, and Notch target genes.
At least eight Ikaros isoforms exist due to alternative splicing. The 2A9-mIkaros antibody has been reported to recognize Ikaros isoform VI (Ik-6), which is the longest and most predominant.
Applications Reported: This 2A9-mIkaros antibody has been reported for use in western blotting.
Applications Tested: This 2A9-mIkaros antibody has been tested by western blot of reduced mouse splenocytes. This can be used at less than or equal to 5 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
DNA-binding protein IKAROS functions in the specification and the maturation of the T lymphocyte. It also interacts with a critical control element in the terminal deoxynucleotidyl transferase promoter as well as with the promoters for other genes expressed during early stages of B-cell and T-cell development.
