Description: The C15.6 antibody reacts with the p40 subunit of mouse interleukin-12 and IL-23. IL-12 is a 70 kDa heterodimeric cytokine composed of two covalently linked chains, p40 and p35, and is secreted by activated B cells and macrophages. IL-23 is a structurally and biologically-related cytokine comprised of the same p40 subunit and a p19 subunit.
For optimal detection of total IL-12 p40 (monomer, heterodimer, homodimer) by ELISA, a combination of purified C15.6 antibody for capture and biotinylated C17.8 antibody for detection is recommended. Fluorochrome conjugated C15.6 antibody is effective for staining intracellular IL-12.
Applications Reported: The C15.6 antibody has been reported for use in ELISA.
Applications Tested: The C15.6 antibody has been tested as the capture antibody in a sandwich ELISA for analysis of mouse IL-12 (p40 monomer or p40 dimerized with p35) or mouse IL-23 (p40 dimerized with p19) in combination with the biotin C17.8 (13-7123) antibody for detection and recombinant mouse IL-12 p70 (14-8121) as the standard. A suitable range of concentrations of this antibody for ELISA capture is 0.25-1 µg/mL. A standard curve consisting of doubling dilutions of the recombinant IL-12 p70 standard over the range of 2000 pg/mL - 15 pg/mL should be included in each ELISA plate.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
IL-12 is an interleukin produced by dendritic cells, neutrophils, macrophages and human lymphoblastoid cells as a result of antigenic stimulation. IL-12 is a heterodimeric cytokine encoded by IL-12A (p35) and IL-12B (p40). IL-23 is a heterodimeric cytokine that shares a subunit with IL-12 (IL-12B) along with an IL-23A (IL-23p19) subunit. Functionally, IL-12 increases the expression of vascular endothelial adhesion molecule-1 on the endothelium, directs leukocytes towards tumors and facilitates the ischemic-hemorrhagic necrosis of the tumor tissue. In comparison, IL-23 plays an important role in stimulating memory T-cells, and is required for the induction, expansion, maintenance and downstream effector functions of Th17 cells, which play a vital role in upregulating neutrophil chemokines and various pro-inflammatory cytokines.
