Description: The C18.2 antibody reacts with the p35 subunit of mouse interleukin-12 (IL-12). Interleukin-12 (IL-12) is a heterodimeric 70 kD (p70) cytokine composed of two covalently linked, glycosylated chains, 40kD (p40) and 35-kD (p35). IL-12 is mainly produced by monocytes, macrophages, and dendritic cells in response to bacterial products such as lipopolysaccharides (LPS), to intracellular pathogens or upon interaction with activated T cells. IL-12 was originally discovered because of its ability to induce interferon-gamma (IFN-g) production, cell proliferation, and cytotoxicity mediated by natural killer cells and T cells. It is now established that IL-12 also plays a key role in the development of Th1 responses, leading to IFN-g and IL-2 production. These cytokines can in turn promote T-cell responses and macrophage activation.
Applications Reported: This C18.2 antibody has been reported for use in cytokine neutralization, and ELISA capture antibody.
Applications Tested: The C18.2 antibody has been tested as the capture antibody in a sandwich ELISA for analysis of bioactive mouse IL-12 p70 in combination with the biotinylated C17.8 (Product # 13-7123-81) antibody for detection and recombinant mouse IL-12 p70 (Product # 14-8121-62) as the standard. A suitable range of concentrations of this antibody for ELISA capture is 1-4 µg/mL. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 2000 pg/mL - 15 pg/mL should be included in each ELISA plate.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Interleukin 12A is a novel signaling cytokine belonging to the Interleukin-6 protein superfamily. Interleukin-12 alpha chain precursor (IL-12A) is a 35-kD subunit of disulfide-linked heterodimer of cytokine IL-12 and has a broad array of biological activities. Reports suggest that IL-12A can act as a growth factor for activated T and NK cells and enhance the lytic activity of NK/lymphokine-activated Killer cells. IL-12A stimulates the production of IFN-gamma by resting PBMC as well as T-cell-independent induction of interferon (IFN)-gamma, and helps in the differentiation of both Th1 and Th2 cells. It is also involved in positive regulation of tyrosine phosphorylation of Stat4 protein. The signaling process of IL-12A in innate immunity requires nitric oxide synthase 2A. Along with EBI3, IL-12A forms a novel heterodimeric cytokine, designated as interleukin-35 (IL-35), that may be specifically produced by T reg cells and is required for maximal suppressive activity.
