Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human IL-16. Anti-Human IL-16-specific antibody was purified by affinity chromatography employing an immobilized Human IL-16 matrix.
Sandwich ELISA: To detect Human IL-16 by sandwich ELISA (using 100 µL/well antibody solution) a concentration of 0.5-2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human IL-16 (500-P06Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human IL-16.
Western Blot: To detect Human IL-16 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human IL-16 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
The protein encoded by IL-16 is a pleiotropic cytokine that functions as a chemoattractant, a modulator of T cell activation, and an inhibitor of HIV replication. The signaling process of this cytokine is mediated by CD4. The product of IL-16 undergoes proteolytic processing, which is found to yield two functional proteins. The cytokine function is exclusively attributed to the secreted C-terminal peptide, while the N-terminal product may play a role in cell cycle control. Caspase 3 is reported to be involved in the proteolytic processing of this protein. Alternate splicing results in multiple transcript variants.
