Description: The WU24A4P monoclonal antibody reacts with human interleukin (IL)-17F homodimer (IL-17FF). This antibody specifically reacts with this homodimer and has no reactivity for the IL-17AF heterodimer. IL-17FF is a 34 kDa member of the IL-17 family, which includes IL-17A, B, C, D, and E (IL-25). Within this family, IL-17A and IL-17F are produced preferentially by T helper 17 (Th17) cells, with other members produced more widely. IL-17F and IL-17A have many overlapping functions, but knockout studies have shown each to have independent functions as well. In vitro cultured Th17 cells have been shown to express IL-17FF and IL-17AA homodimers and IL-17AF heterodimers depending on the culture conditions and their differentiation state. Together, these three dimers signal through the IL-17RA/IL-17RC receptor complex to mediate immune responses at mucosal interfaces and are found at lesion sites in inflammatory bowel disease, asthma, atopic dermatitis and rheumatoid arthritis.
While other clones with specificity for IL-17F, such as SHLR17, react with the IL-17F monomer in the context both IL-17FF and IL-17AF dimers, clone WU24A4P reacts specifically with IL-17FF homodimer and does not detect IL-17AF heterodimer.
Applications Reported: This WU24A4P antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested: This WU24A4P antibody has been pre-titrated and tested by intracellualr staining and flow cytometric analysis of Th17-polarized normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to Best Protocols: Protocol A: Two step protocol for (cytoplasmic) intracellular proteins located under the Resources Tab online. This can be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
eFluor® 660 is a replacement for Alexa Fluor® 647. eFluor® 660 emits at 659 nm and is excited with the red laser (633 nm). Please make sure that your instrument is capable of detecting this fluorochome.

Interleukin 17F is a cytokine belonging to the IL-17 family that is produced by inflammatory cells such as activated T cells, mast cells, and basophils. IL-17F is involved in allergic airway inflammation, and can induce several cytokines, chemokines, and adhesion molecules in bronchial epithelial cells, vein endothelial cells, fibroblasts, and eosinophils. IL-17F may be secreted as a homodimer, or a heterodimer with IL17A. It acts by binding to the type I receptor, IL-17R, aiding recruitment of monocytes and neutrophils at the site of inflamation by increasing chemokine production.
