Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human MMP-3. Anti-Human MMP-3-specific antibody was purified by affinity chromatography employing an immobilized Human MMP-3 matrix.
Sandwich ELISA: To detect Human MMP-3 by sandwich ELISA (using 100 µL/well), a concentration of 0.5-2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human MMP-3 (500-P324BT) as a detection antibody, allows the detection of at least 2000-4000 pg/mL of Recombinant Human MMP-3.
Western Blot: To detect Human MMP-3 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. When used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human MMP-3 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
MMP3 (stromelysin-1) belongs to the peptidase M10A subfamily. It is secreted in its pro-protein form, and is activated by extracellular proteinase cleavage. MMP3 is capable of binding to zinc and calcium ions, and has metallo-endopeptidase activity. The active enzyme degrades collagen, gelatin, fibronectin, and laminin. It is involved in normal cellular processes such as wound repair and tissue remodeling. Mutations in the gene can result in coronary heart disease 6.
