PKP3 Antibody — High K8 levels in the PKP3 knockdown clones are due to a decrease in phosphorylation. A. Protein extracts from the vector control (pTU6) and PKP3 knockdown clones (S9 and S10) were resolved on SDSPAGE gels and Western blots performed with the indicated antibodies. B. S9 cells were transfected with either dsRed or the shRNA resistant dsRed PKP3 3.7R cDNA. 48 hours post transfection cells were stained with antibodies to K8 (green) and visualized by confocal microscopy. MA1-06511 detects plakophilin-3 in human, mouse, canine cells.

MA1-06511 has sucessfully been used in Western blotting, immunoprecipitation, immunocytochemistry and immunohistochemistry procedures. By Western blot it detects a ~87 kDa band representing Plakophilin-3.

The MA1-06511 immunogen is a synthetic peptide corresponding to residues F(570) T P Q S R R L R E L P L A A D A L T F(589) of human plakophilin-3.

Target Information

Plakophilin-3 is an Armadillo-like protein present in nuclei and desmosomes of epithelial cells. The expression pattern of this protein seems to be largely restricted to epithelial cell types. Plakophilin-3 can be detected along cell borders in a punctuate staining pattern typical for desmosomal proteins. In addition to the desmosomal immunolocalization, immunostaining was observed as bright nuclear speckles. Thus, like plakophilin-1 and-2, plakophilin-3 displays a dual intracellular localization in the desmosomal plaque and in the cell nucleus, and therefore is probably involved in signal transduction pathways between the plasma membrane and the nucleus. The human protein has a predicted molecular mass of ~87 kDa.
