Detection of human SF3B5 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-SF3B5 antibody A305-587A-M (lot A305-587A-M-1) used for IP at 8 µl per reaction. SF3B5 was also immunoprecipitated by rabbit anti-SF3B5 antibody A305-586A-M. Target Information
SF3B5 is a protein coding gene involved in pre-mRNA splicing as a component of the splicing factor SF3B complex, a constituent of the spliceosome. SF3B complex is required for 'A' complex assembly formed by the stable binding of U2 snRNP to the branchpoint sequence (BPS) in pre-mRNA. Sequence independent binding of SF3A/SF3B complex upstream of the branch site is essential, it may anchor U2 snRNP to the pre-mRNA.
