This antibody neutralizes the biological activity of recombinant mouse Thrombopoietin/Tpo. This antibody will also neutralize the activity of recombinant human (rh) Thrombopoietin/Tpo, although 25 times the amount of Ig is required. In direct ELISAs less than 15% cross-reactivity with rhTpo is observed.
Reconstitute at 0.2 mg/mL in sterile PBS.
Endoxin level is <0.10 EU per 1 µg of the antibody by the LAL method.
Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT signaling pathways that leads to the proliferation and differentiation of megakaryocytes and, ultimately, to the production of platelets (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
