Description: This MR54 monoclonal antibody reacts with human Trop-2, which is also known as Tumor-Associated Calcium Signal Transducer 2 (TACSTD2) and EGP-1. This type I transmembrane glycoprotein is expressed on trophoblast (syncytiotrophoblast and cytotrophoblast), epithelial, and most human carcinoma cells. Trop-2 plays a role in calcium-mediated signaling. Trop-2 has also been reported as an oncogene, with expression being correlated with tumorigenesis, metastasis, and anchorage-independent growth.
Applications Reported: This MR54 antibody has been reported for use in flow cytometric analysis, immunoprecipitation, western blotting, immunohistochemical staining of formalin-fixed paraffin embedded (FFPE) tissue sections (IHC-P), ELISA, and immunocytochemistry (ICC).
Applications Tested: This MR54 antibody has been tested by flow cytometry and immunocytochemistry on MCF7 cells as well as by immunohistochemistry of formalin-fixed paraffin embedded tissue using either low or high pH antigen retrieval methods. Use of high pH antigen retrieval results in better staining intensity. This can be used at less than of equal to 5 µg/mL. Is is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
TROP2 is a carcinoma-associated antigen defined by the monoclonal antibody GA733. This antigen is a member of a family including at least two type I membrane proteins. It transduces an intracellular calcium signal and acts as a cell surface receptor. Mutations of its gene result in gelatinous drop-like corneal dystrophy, an autosomal recessive disorder characterized by severe corneal amyloidosis leading to blindness.
