Immunofluorescence analysis of Villin was performed using 70 confluent log phase Caco-2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is colon, small intestine or kidney tissue.
Target Information
Villin is part of cytoskeleton associated with the microvillar actin core bundle of gastrointestinal and renal brush border. It can interact with actin in a Ca2+ and phosphoinositide-regulated manner. Villin is a very specific marker for adenocarcinomas of the gastrointestinal tract and the pancreas. It is also expressed in some Merkel cell tumor and adenocarcinoma of the lung, prostate, ovarian and kidney.
