Store at 4° C or -20°C. The working antibody solution is stable for at least 7 days at 4°C. Precautions should be taken for storage for longer periods. Problems of long term stability may occur with highly diluted solutions. No other preservative agent has been added to the present formulation. For long storage purposes in solution the addition of sodium azide to 0.02 % is advised with the appropriate precautions of use.
Precipitation assays: In immunoelectrophoresis in agarose-plates use 2 µL human plasma or equivalent against 120 µL antiserum. In double radial immunodiffusion use a rosette arrangement with 10 µL antiserum in 3 mm diameter center well and 2 µL plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells. In electroimmunodiffusion (Laurell) use a concentration of 1% of the antiserum in the gel.
Species Reactivity: The antiserum does not cross-react with any other Human plasma proteins as tested in gel-diffusion techniques. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins, since homologous proteins of different species frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.
Reconstitute the lyophilized antiserum by adding 1 mL sterile distilled water.
Trypsin is a 24 kDa enzyme of the serine proteinase family. It is produced in the pancreas as an inactive precursor, trypsinogen, but the active enzyme is located in the gastrointestinal tract where it degrades proteins to large peptides. Trypsin prefers lysine and arginine residues. One of the substrates for trypsin is chymotrypsinogen which is cleaved to produce active chymotrypsin. High levels of immunoreactive trypsin in the bloodstream can indicate pancreatic malfunction and this indicator is used to screen for and diagnose Cystic Fibrosis.
