The stability and bioavailability of an active pharmaceutical ingredient (API) in a drug is key to that drug achieving its desired effect. Supersaturating drug delivery systems, such as amorphous solid dispersions, is one approach to improve solubility.
In this webinar, the speaker discusses increasing the aqueous solubility of orally administered poorly water-soluble drugs. This is done by creating a supersaturation state in the gastrointestinal lumen, thus improving bioavailability.

The search for effective ways to increase the solubility of drugs is constant.
Hydroxypropyl methylcellulose acetate succinate (HPMCAS) can keep poorly water-soluble drugs in a supersaturated state after dissolution, so it's become a more popular carrier for amorphous solid dispersions (ASDs). Yet HPMCAS has mostly been limited to ASDs prepared by spray drying. It would be preferable to use the solvent-free method of hot-melt extrusion (HME) to prepare ASDs, but the high temperatures required can degrade the polymer.
This webinar reviews the use of solid surfactants to reduce complex viscosity (and thus the processing temperature) of HPMCAS-based ASDs. The speaker presents a study where surfactant-drug-polymer mixtures were successfully extruded, and the resulting ASDs were developed into tablets. The analysis paves the way for greater use of HME in producing HPMCAS-based ASDs.
