The Human Heat Shock Protein 27(Hu HSP27) ELISA quantitates Hu HSP27 in human serum, plasma, cell lysates, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu HSP27.
Principle of the method
The Human HSP27 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

In response to adverse changes in their environment, cells from many organisms increase the expression of a class of proteins referred to as heat shock or stress proteins. HSPB1 (heat shock protein beta-1 or HSP27) is a small heat shock protein which functions as a molecular chaperone that maintains denatured proteins in a folding-competent state. It plays a role in stress resistance and actin organization. Through its molecular chaperone activity, HSP27 regulates numerous biological processes including the phosphorylation and the axonal transport of neurofilament proteins. Mutations in the gene can result in Charcot-Marie-Tooth disease 2F and Neuronopathy distal hereditary motor 2B.
