The Human Matrix MetalloProteinase 13 (Hu MMP13) ELISA quantitates Hu MMP13 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu MMP13.
Principle of the method
The Human MMP13 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

MMP13 (collagense 3) plays a role in the degradation of extracellular matrix proteins including fibrillar collagen, fibronectin, TNC, and ACAN. MMP13 cleaves tripal helical collagens, including type I, II, and III collagen - but has the highest activity with soluble type II collagen. It can degrade collagen type IV, XIV, and X. MMP13 plays a role in wound healing, tissue remodeling, cartilage degradation, bone development, bone mineralization, and ossification. It is required for normal embryonic bone development and ossificaiton. Mutations affecting the gene can lead to spondyloepimetaphyseal dysplasia Missouri type, metaphyseal anadysplasia 1, and metaphyseal dysplasia Sphar type.
