The Mouse Fractalkine/CX3CL1 ELISA quantitates Ms Fractalkine in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms Fractalkine.
Principle of the method
The Mouse Fractalkine solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Chemokines are a family of proteins associated with the trafficking of leukocytes in immune surveillance and inflammatory cell recruitment. They are classified based on the positions of key cysteine residues. CX3CL1 is a CX3C chemokine known to induce adhesion and migration of leukocytes mediated by a membrane-bound and soluble form respectively. Recent experiments have shown that CX3CL1 can suppress the production of nitrous oxide, interleukin-6, and TNF-a in activated microglia and neuronal cells, suggesting that it may act as an intrinsic inhibitor against neurotoxicity by activated microglia. Its receptor, CX3CR1, also functions as a co-receptor for HIV-1 and HIV-2 envelope fusion and virus infection, which can be inhibited by CX3CL1.
