The Rat Tissue Inhibitor of Matrix MetalloProteinase 1-Cell Lysates (Rt TIMP-1) ELISA quantitates Rt TIMP-1 in rat cell or tissue lysates. The assay will exclusively recognize both natural and recombinant Rt TIMP-1.
Principle of the method
The Rat TIMP-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

TIMP1 (metalloproteinase inhibitor 1) is a metalloproteinase inhibitor that functions by forms one-to-one complexes with target metallproteinases, such as collagenases, and irreversible inactivates them by binding to their catalytic zinc cofactor. TIMP1 acts on MMP1, MMP2, MMP3, MMP7, MMP8, MMP9, MMP10, MMP11, MMP12, MMP14, and MMP16. It also functions as a growth factor that regulates cell differenation, migration, and cell death. TIMP1 activates cellular signaling cascades via CD63 and ITGB1.
