Reconstitution: We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute lyophilized human GDNF in sterile, distilled water to 0.1-0.5 mg/mL. These stock solutions should be apportioned into working aliquots and stored at ≤-20°C. Further dilution should be made in medium or buffered solution containing carrier protein, such as PBS with 0.1% BSA. Storage: Lyophilized human GDNF should be stored at 2°C to 8°C, preferably desiccated. Store reconstituted human GDNF at ≤-20°C (not in a frost-free freezer). Keep freeze-thaw cycles to a minimum.
GDNF is a glycosylated, disulfide-bonded homodimer molecule. It was first discovered as a potent survival factor for midbrain dopaminergic neurons and was then shown to rescue these neurons in animal models of Parkinson's disease. GDNF is about 100 times more efficient survival factor for spinal motor neurons than the neurotrophins. GDNF is a neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.
