Mature human IL-2 was expressed in Pichia pastoris, X-33. GenBank Accession # NP_000577, amino acids 21 - 153. For protein verification, the expressed protein was N-terminal sequenced and begins with the following 10 amino acids: APTSSSTKKT. Purity is >95% by SDS-PAGE analyses.
Activity: The biological activity was determined by measuring the proliferation of mouse HEK-Blue cells.
Use: For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments.
Reconstitution and Dilution: Store lyophilized material at 4ºC. Reconstitute in 1 mL of distilled or deionized water. Dilute in a buffer-containing carrier protein, aliquot, and store at or below -70ºC, avoiding multiple freeze-thaw cycles. PBS with 1% BSA or a neutral media containing <10% FCS are suitable buffers.
Interleukin 2 (IL-2) is an immuno-modulatory cytokine that is important for the proliferation of activated T cells, differentiation of B cells, natural killer cells, monocytes and macrophages. IL-2 signals through the IL-2 receptor (IL-2R), a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL-4) and interleukin 7 (IL-7). The expression of the IL-2 gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a gene similar to IL-2 in mice leads to an ulcerative colitis-like disease that suggests an essential role of this gene in the immune response to antigenic stimuli.
