Mature rhesus macaque IL-18 was expressed in Pichia pastoris, X-33, GenBank Accession # AAK13416, amino acids 37 - 193. For protein verification, the expressed protein was N-terminal sequenced and begins with the following 10 amino acids: YFGKLESKLS. Purity is >95% by SDS-PAGE analyses.
Activity: The biological activity of recombinant rhesus macaque IL-18 was determined by its ability to induce IFNg production from KG-1 cells in the presence of recombinant mouse IL-12. 270 pg/mL and 470 pg/mL of IFNg are induced by 20 ng/mL and 40 ng/mL of recombinant rhesus macaque IL-18, respectively.
Use: For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments.
Reconstitution and Dilution: Store lyophilized material at 4ºC. Reconstitute in 0.5 mL of distilled or deionized water. Dilute in a buffer-containing carrier protein, aliquot, and store at or below -70ºC, avoiding multiple freeze-thaw cycles. PBS with 1% BSA or a neutral media containing <10% FCS are suitable buffers.
The protein encoded by this gene is a proinflammatory cytokine. This cytokine can induce the IFN-gamma production of T cells. The combination of this cytokine and IL12 has been shown to inhibit IL4 dependent IgE and IgG1 production, and enhance IgG2a production of B cells. IL-18 binding protein can specifically interact with this cytokine, and thus negatively regulate its biological activity.
