Announcing EVOS FL Auto 2 Software Revision 2.0.2094.0
Revision 2.0.2094.0 introduces new and enhanced features, including improved autofocus in fluorescence and transmitted light, the ability to control stage acceleration and speed. It also introduces the EVOS Analysis application that provides tools for annotations and measurements, histograms and cell analysis. It also includes bug fixes, including correction of a a tile-stitching artifact.

New features
Improved autofocus
- Improved autofocus speed and accuracy in fluorescence and transmitted light to make it easier to focus.
Measurements and annotations
- Provides users with a range of line and area tools for highlighting and measurements of objects in an image, plus the ability to add annotations.
Histograms
- Enables users to view intensity information across the field or annotated shape with a histogram.
Auto cell count
- Obtain total count of objects captured in fluorescence, e.g. nuclei stained with Hoechst, by selecting target and background. Calculations can be refined with undesired objects gated out based on size, intensity, or circularity. Apply to a single field or to a multi-well plate using batch analysis.
Confluence
- Automatically measure and document the percent confluence, (the area covered by adherent cells) in a transmitted light image by selecting cells and background. A sensitivity slider can be used to refine the results. Use of saved settings can provide consistent cell passaging and increase reproducibility of downstream cell-based assays. Use it for a single field or in batch mode for multiple fields and wells.
Transfection efficiency
- Calculate transfection efficiency based on a user-adjustable threshold of positive pixels in the fluorescence channel after determining confluence. Percent transfection is based on the ratio of pixels in fluorescence compared to those in confluence. Can be applied to a single field or to multiple fields using batch analysis. This tool helps assess and optimize transfection conditions and cell assay readiness.
10x genomics slide support
- Vessel maps are now available for the Visium Tissue Optimization slide and Visium Gene Expression slide from 10X Genomics.
Stage control
- Gives users the ability to reduce stage acceleration and deceleration to facilitate less disruption of cells in samples with suspension or weakly adherent cells, e.g., spheroids, for increased reproducibility and improved data quality.
Celleste software launch button
- A button in the Review tab enables the launch of Celleste Image Analysis software for seamless analysis of images captured on EVOS FL Auto 2.
Bug fixes
Tile-stitching
- An artifact that introduces pixel shifts during tile-stitching of scanned images has been fixed
Pseudocolor settings
- Pseudocolor changes are now retained and applied through different applications, including when moving to new vessel maps
Scan and saving
- Images acquired during a scan are now retained even if the scan is cancelled early if Save is selected
Installation Instructions
Software Update (Rev 2.0.2094.0)
IMPORTANT! Restart your system prior to updating to optimize the update process.
Update from USB
- Insert a fresh USB drive with at least 240 MB available space.
- Right-click the “Download” link for Software Update.
- Select “Save Link as”.
- Select the USB Drive and press Save.
- Plug the USB flash drive into the M7000 computer.
- Locate the update file from your USB drive in Windows Explorer and copy it to the hard drive.
- Double-click the update file from the hard drive to install the update.
- A WindowsTM dialog box will pop up. Click Next to proceed with the software update.
- The screen will display the update progress. When the update is complete, click OK.
- Shut down the computer (Windows Start -> Power icon -> Shut down).
- Power up the computer and launch the M7000 software.
The installation process will take approximately 5-10 minutes to complete on your instrument.
