Potential-dependent accumulation of the cationic dye in mitochondria results in a relatively weak fluorescence signal due to self-quenching (upper panel). Dissipation of the mitochondrial membrane potential by the uncoupler FCCP is marked by increasing fluorescence (middle panel) and subsequent redistribution of the dye throughout the cell (lower panel).
