Description: The X2/45-12 monoclonal antibody reacts with the human IL-4 receptor alpha chain. IL-4 receptor alpha chain (CD124) is crucial for the binding and signal transduction of IL-4. Both IL-4 and IL-13 use the IL-4 receptor alpha chain as their receptor component and activate many common signal pathways.
Applications Reported: X2/45-12 has been reported for use in flow cytometric analysis. X2/45-12 has also been reported in neutralization of human IL-4 function.
Applications Tested: The X2/45-12 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 1 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Storage and handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: Greater than 90%, as determined by SDS-PAGE.
Endotoxin Level: Less than 0.001 ng/µg antibody, as determined by LAL assay.
Aggregation: Less than 10%, as determined by HPLC.
IL4 Receptor alpha (IL4RA) is the alpha chain of the interleukin 4 receptor which binds to both interleukin 4 and interleukin 13 to regulate IgE production, chemokine and mucus production at sites of allergic inflammmation. The IL4 response is involved in promoting Th2 differentiation. The secreted extracellular domain of IL4R alpha, called sIL4R alpha, can inhibit IL4-mediated cell proliferation and IL5 upregulation by T-cells. It has no signaling abilities. Allelic variations in the IL4RA gene have been associated with atopy, a condition that can manifest itself as allergic rhinitis, sinusitus, asthma, or eczema.
