Description: The DL-101 monoclonal antibody reacts with human CD138, also known as syndecan-1. CD138 is a transmembrane protein containing chondroitin sulfate and heparin sulfate moieties responsible for binding to the extracellular matrix components. CD138 is not expressed by mature B cells but is present on pre-B cells and the finally differentiated plasma cells.
Applications Reported: This DL-101 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This DL-101 antibody has been pre-diluted and tested by flow cytometric analysis of U266 cells. This may be used at 5 µL (1.0 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm. We recommend using a 450/50 bandpass filter, or equivalent. Please make sure that your instrument is capable of detecting this fluorochrome.
When using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer for more information.

CD138 (syndecan 1) is a transmembrane proteoglycan that can bind a variety of cytokines and modulate their activity, as well as the activity of extracellular matrix components and influence many developmental processes. CD138 is expressed mainly in differentiating keratinocytes and is transiently upregulated in all layers of the epidermis upon tissue injury. It is also highly expressed on plasma cells and can be detected even on fibroblasts, vascular smooth muscle cells and endothelial cells. Up-regulation and down-regulation of CD138 on the cell surface often correlates with the gain of cancerous characteristics. Serum levels of the shedded soluble sCD138 are used as a prognostic factor of cancerogenesis.
