Description: The monoclonal antibody 20C8 recognizes human Snail1, a 29 kDa zinc finger transcription factor. Snail1 and Slug both belong to a family of transcription factors that are responsible for inducing epithelial to mesenchymal transition (EMT) during normal organ development and tumor progression. Snail1 expression is localized to both the cytoplasm and nucleus and functions as a transcriptional repressor of E-cadherin, claudins, occludin, and desmoplakin. During embryonic development, Snail1 is localized to the mesoderm, while in the adult, expression is inducible and is usually limited to activated fibroblasts and some mesenchymal cells. Overexpression of Snail1 has been found in carcinomas and fibrosarcomas and may be used as an early marker for EMT.
Applications Reported: This 20C8 antibody has been reported for use in immunohistochemical staining of formalin-fixed paraffin embedded (IHC-P) tissue, ELISA, immunoprecipitation, western blotting, immunocytochemistry, and intracellular staining followed by flow cytometric analysis.
Applications Tested: This 20C8 antibody has been tested by immunocytochemistry on fixed and permeabilized MCF7 cells and by immunohistochemistry on formalin-fixed paraffin embedded human infiltrating ductal carcinoma. This antibody can be used at less than or equal to 20 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Zinc finger protein SNAI1 (SNAIL)is a zinc finger transcriptional repressor which down regulates the expression of the ectodermal gene within the mesoderm and functions to form the mesoderm and neural crest. It is reported that SNAI1 acts as an important factor of tumor invasion as evidenced by its role in E-cadherin down-regulation and induction of epithelial-mesenchymal transition (EMT). In human breast cancer, the expression of SNAI1 and/or the homologous SNAI2 (Slug) has been associated with E-cadherin repression, local or distant metastasis, tumor recurrence, or poor prognosis in different tumor series. It is also reported that Snail 1 protein in the stromamay be anew putative prognosis marker for colon tumors.
