To address the HIV surveillance research needs of the global community, the Applied Biosystems HIV-1 Genotyping Kit with Integrase offers broad genotyping coverage of HIV-1 Group M subtypes from extracted viral RNA from plasma and dried blood spot (DBS) samples to detect resistance to protease inhibitors, nucleoside reverse-transcriptase inhibitors, non-nucleoside reverse-transcriptase inhibitors, and integrase inhibitors.

This HIV-1 drug resistance research use only (RUO) kit harnesses gold-standard Sanger sequencing technology to amplify and reliably sequence the diverse and rapidly evolving HIV-1 virus.

This product employs assays for HIV-1 genotyping licensed from the U.S. Centers for Disease Control and Prevention.

Survey HIVDR mutations with the CDC

Having an accurate and sensitive test for HIV-1 genetic variants that resist common antiretroviral therapeutics is critical for epidemiologic and genetic surveillance studies. Here, a scientist from the United States Centers for Disease Control (CDC) discusses the CDC’s experience investigating the HIV-1 Genotyping Kit with Integrase.

The HIV-1 Genotyping Kit with Integrase features:

  • A standardized, cost-effective assay in a scalable workflow
  • A total workflow of about 20 hours for processing extracted viral RNA from EDTA plasma or DBS to results, with total hands-on time of ~4.5 hours
  • The ability to detect mutations in the protease, reverse transcriptase, and integrase regions of the HIV-1 pol gene in subtypes A, B, C, D, F, G, CRF01_AE, CRF02_AG, and CRF06_CPX
  • Easy-to-read sequencing results with software for assembly and interpretation at no additional charge
  • Compatibility with a pre-analytical solution for nucleic acid extraction from EDTA plasma and dried blood spots

Assay workflow

Nucleic acids from plasma or dried blood spot (DBS) samples are extracted. Extraction can be performed with the MagMAX Viral/Pathogen Nucleic Acid Isolation Kit for HIV-1 or other methods.

After sample processing and RNA extraction, samples undergo an initial reverse transcription + PCR, followed by a second nested PCR step for mutation targets of interest. The HIV-1 Genotyping Kit includes the RT-PCR master mix, the nested PCR master mix, and necessary positive and negative controls that allow for a simple, streamlined workflow as you move into your cleanup and sequencing steps.

Once you confirm and clean up your targeted PCR product, the remainder of the HIV-1 Genotyping Kit with Integrase is used for cycle sequencing. Included in this kit are the forward/reverse pre-formulated sequencing mixes for the targets of interest.

Free access to Hyrax Biosciences' online platform, Exatype™, is provided with the purchase of HIV-1 Genotyping Kits. This easy-to-use application allows for:
