Alkaline Phosphatase staining of iPSC colonies

In a and b, 200,000 primary human fibroblasts were reprogrammed with CytoTune™ -iPS Sendai Reprogramming Kit (a) and Lentivirus (b). Twenty-eight days post reprogramming, ~245 colonies were observed in the CytoTune™ reprogramming condition while the lentivirus reprogramming condition generated ~10 colonies. In c and d, 50,000 primary human fibroblasts were reprogrammed with CytoTune™ -iPS Sendai Reprogramming Kit (c) and mRNA (d). Twenty-eight days post reprogramming, ~130 iPSC colonies were observed in the CytoTune™ reprogramming condition while 11 iPSC colonies were observed in the mRNA reprogramming condition.
