1D Electrophoresis Chamber Systems—General

Our Original Bolt™ Bis-Tris Plus Mini gels (Cat. No. BGxxxxxBOX, discontinued as of December 31, 2014) can only be run in the Bolt™ Mini Gel Tank (discontinued as of December 31, 2014, and will be offered until inventory is depleted).

Our New Bolt™ Bis-Tris Plus Mini gels (Cat. No. NWxxxxxBOX), as well as our Novex™ Mini gels and NuPAGE™ Mini gels can be run using the Mini Gel Tank, or XCell SureLock™ Mini-Cell. To run these gels using the Bolt™ Mini Gel Tank (discontinued as of December 31, 2014), upgrading of the tank is necessary by replacing the black 10.5 cm cassette clamp cam handles with gray 10 cm cassette clamp cam handles (Cat. No. A26732, Cassette Clamp Cam Handle Set). Instructions for replacement of the cam handles can be found here or in this video.

Our Midi gels can be run using the XCell4 SureLock™ Midi-Cell.

Yes, the 10 cm x 10 cm x 7 mm cassette Precise Tris-Glycine gels are compatible with the XCell SureLock™ Mini Cell and the Mini Gel Tank.

The Precise™ Tris-HEPES gels are compatible with the XCell SureLock™ Mini Cell or Mini Gel Tank when used with adaptor plates.

Note: Two adaptor plates are required when running just one gel and one adaptor plate is required when running two gels using the XCell SureLock™ Mini Cell or Mini Gel Tank.

We used to offer the XCell6™ MultiGel unit for simultaneous running of up to 6 mini gels, but it has been discontinued.

1D Electrophoresis Chamber Systems—XCell SureLock™ Mini-Cell

The XCell SureLock™ Mini-Cell includes the lid, gel tension wedge, lower buffer chamber, buffer core, buffer dam, and gel knife.

Our New Bolt™ Bis-Tris Plus Mini gels (Cat. No. NWxxxxxBOX), as well as our Novex™ Mini gels and NuPAGE™ Mini gels can be run using the XCell SureLock™ Mini-Cell.

The XCell II™ Mini-Cell has been discontinued and can be upgraded to an XCell SureLock™ Mini-Cell with the purchase of the SureLock™ Retrofit kit for XCell II™ Mini-Cell, Cat. No. EI0020 (consists of a SureLock™ mini-cell lid, gel tension wedge, and buffer dam).

We recommend washing the XCell SureLock™ Mini-Cell with a mild detergent and rinsing with deionized water after each use.

You can try the standard recommendation for cleaning which would be washing the unit with a mild detergent and rinsing with deionized water.

The XCell SureLock™ Mini-Cell is impervious to alcohol, but not compatible with chlorinated hydrocarbons (e.g., chloroform), aromatic hydrocarbons (e.g., toluene, benzene) or acetone.

Our Original Bolt™ Bis-Tris Plus gels (Cat. No. BGxxxxxBOX), discontinued as of December 31, 2014) do not fit in the XCell SureLock™ Mini-Cell. They need to be run in the Bolt™ Mini Gel Tank (discontinued as of December 31, 2014). Our New Bolt™ Bis-Tris Plus Mini gels (Cat. No. NWxxxxxBOX) can be run using the XCell SureLock™ Mini-Cell (or Mini Gel Tank, Cat. No. A25977).

The Mini Gel Tank is not compatible with chlorinated hydrocarbons (e.g., chloroform), aromatic hydrocarbons (e.g., toluene, benzene) or acetone.

Here are our recommendations:

  • When electrophoresis is complete, dispose of the buffer appropriately. Rinse the tank with water to remove residual buffer.
  • Clean the surface of the Mini Gel Tank with a soft non-abrasive, lint-free cloth dampened with water.
  • Do not use harsh detergents or solvents to clean the unit.

We offer the PowerEase™ 90W Power supply that is compatible with the Bolt™ Mini Gel Tank. The PowerEase™ 500 Power Supply (discontinued as of December 31, 2013) is also compatible with the Bolt™ Mini Gel Tank). Novex™ Power Supply Adapters are supplied with the tank for power supplies that are not designed for use with covered or non-retractable power leads.

To run Mini gels with 10 cm gel cassettes using a Bolt™ Mini Gel Tank (without replacement of 10.5 cm cassette clamp cam handles with 10 cm cassette clamp cam handles), please use the instructions provided on Page 22 of the manual.

1D Electrophoresis Chamber Systems—XCell4 SureLock™ Midi-Cell

The XCell4 SureLock™ Midi-Cell includes the lower buffer chamber, lid with cables, two buffer cores, buffer dam, displacement dam, SureLock™ assembly, and gel knife.

Our Midi gels can be run using the XCell4 SureLock™ Midi-Cell.

Yes, the XCell4 SureLock™ Midi-Cell is designed to perform simultaneous vertical electrophoresis of 1–4 midi-gels. Use the XCell4 Displacement Dam when you wish to run less than 3 gels in the XCell4 SureLock™ Midi-Cell. The Displacement Dam is placed in the Lower Buffer Chamber instead of a second Buffer Core. Do not add any buffer or water to the Displacement Dam. However, if water or buffer is accidentally added to the Displacement Dam, it does not affect the electrophoresis run.

No. While the NuPAGE™ Novex™ Midi Gels are compatible with the Bio-Rad™ Criterion system (when the specific adapter is used), the Bio-Rad™ Criterion gels will not work with the XCell4 SureLock™ Midi-Cell.

Note: Our Mini gels are not compatible with the Bio-Rad™ Mini-PROTEAN™ apparatus.

The XCell4 SureLock™ Midi-Cell is impervious to alcohol, but not compatible with chlorinated hydrocarbons (e.g., chloroform), aromatic hydrocarbons (e.g., toluene, benzene) or acetone.

Use the XCell4 Displacement Dam when you wish to run less than 3 gels in the XCell4 SureLock™ Midi-Cell. The Displacement Dam is placed in the Lower Buffer Chamber instead of a second Buffer Core. Do not add any buffer or water to the Displacement Dam. However, if water or buffer is accidentally added to the Displacement Dam, it does not affect the electrophoresis run.

The XCell4 SureLock™ Assembly or CAM is similar to a gel tension wedge. The positive locking action of the XCell SureLock™ Assembly ensures trouble-free, leak-free electrophoresis of protein gels. Move the lever on the XCell SureLock™ Assembly to a locked position to generate an even horizontal force which seals the gel cassette/Buffer Core assembly firmly into position in the Lower Buffer Chamber. The locked and unlocked positions are marked on the XCell SureLock™ Assembly for easy, intuitive assembly.

We do not recommend using the gel tension wedge that is supplied with the XCell SureLock™ Mini-Cell for the XCell4 SureLock™ Midi-Cell as it is specifically designed for the Mini-Cell and will not fit correctly in the Midi-Cell.

The XCell4 SureLock™ Midi-Cell should be used with an external DC power supply designed for electrophoresis. This power supply must:

  • Be isolated from the ground so that the DC output is floating (safety requirement)and must be equipped with No Load or Open Circuit protection
  • Be able to operate at 200 V

If you would like to simultaneously run 4 gels using the XCell4 SureLock™ Midi-Cell, we recommend using the ZOOM™ Dual Power Supply. The Low Voltage/High Current outlets can handle the voltage and current of 4 NuPAGE™ Bis-Tris gels (Voltage 200 V, Current ~800 mA).

The PowerEase™ 90W Power Supply and PowerEase™ 500 Power Supply (discontinued as of December 31, 2013) can be used with the XCell4 SureLock™ Midi-Cell but the current limit of 500 mA will be reached with 2 gels (2 x ~200 mA max current at 200V). If 4 gels are run in the XCell4 SureLock™ Midi-Cell with these power supplies, the current will be maxed out and the run will slow down. It will take approximately twice as long to run the gels.

Note: To allow connection of the covered power leads of the XCell4 SureLock™ Midi-Cell with certain power supplies that are not designed for use with covered or non-retractable power leads, use Novex™ Power Supply Adapters available separately (Cat. No. ZA10001).

We recommend washing the unit with a mild detergent and rinsing with deionized water after each use.

1D Electrophoresis Power Supplies—PowerEase™ 300W Power Supply (discontinued)

The PowerEase™ 300W Power Supply is a fully programmable, microprocessor-controlled power supply designed for high-throughput gel electrophoresis. It has a straight-forward, intuitive interphase and includes the following features:

  • Constant voltage, current, or power settings
  • Built-in timer for walk-away gel electrophoresis
  • Up to 10 custom programs with 10 steps each
  • Four sets of output jacks that are compatible with most electrophoresis devices

The PowerEase™ 300W Power Supply accommodates the running and transferring of 8 Mini gels, or 16 Midi gels.

The PowerEase™ 300W Power Supply includes the following components:

  • PowerEase™ 300W Power Supply
  • Extra fuse
  • Power cord

Note: Please use the power cord approved for your country/region. North America customers should use the Type B power cord; Japanese customers should use the JIS 8303 power cord; Chinese customers should use the GB 15934-2008 power cord; Australia/New Zealand customers should use the Type I power cord.

The product specifications for the PowerEase™ 300W Power Supply are listed on Page 18 of the manual.

For best results, we do not recommend using the PowerEase™ 300W Power Supply at its maximum electrical load limits. Variations in buffer conditions can result in exceeding the power supply’s maximum voltage, current, or power output capacity and produce undesirable variations in performance.

When using the PowerEase™ 300W Power Supply with 1–2 proteins gels or blots, we recommend using the Constant Voltage mode for optimal performance.

The PowerEase™ 300W Power Supply is capable of operating at limiting voltage, limiting current, or limiting power. However, we recommend operating it at limiting voltage for most applications as it provides the following advantages:

  • For most electrophoresis methods, resistance increases throughout the run. Under limiting voltage, current and power decrease throughout the run, providing an improving margin of safety over time.
  • The same voltage setting can be used regardless of the number or thickness of gels being electrophoresed.

Note: 

With the current limiting setting, the voltage will increase as resistance increases to satisfy Ohm’s law (V=IR). If no voltage limit is set and a local fault condition occurs, such as a poor connection, very high local resistance may cause the voltage to increase to the maximum capacity of the power supply. This may lead to local overheating and damage to the electrophoresis cell or create unsafe conditions. When operating under constant current conditions, we recommend setting a voltage limit on the power supply at or slightly above the maximum expected voltage.

Power is a function of voltage and current P=IV. For a typical gel run, the current will decrease causing the voltage to increase when run at constant power. The power limiting function may be used when running sequencing gels to remove the ammonium persulfate from the wells and to heat the gel to an optimal temperature for DNA separations.

The PowerEase™ 300W Power Supply requires no periodic maintenance with the exception of an occasional dry wipe-down of the instrument.

During NativePAGE™ electrophoresis, it is common for the current to drop below 1 mA. The PowerEase™ 300W Power Supply will register this as a “No Load” error and automatically shut off, resulting in the stopping of the gel run. Since there is no way to disable/turn off the “Load Check” feature in the PowerEase™ 300W Power Supply, we do not recommend using it for Native-PAGE electrophoresis.

1D Electrophoresis Power Supplies—PowerEase™ 90W Power Supply (discontinued)

The PowerEase™ 90W Power Supply is a microprocessor-controlled power supply designed specifically for mini-gel electrophoresis. The straight-forward, intuitive interface makes the powering of DNA and protein gel runs a simple and easy process. In addition, the PowerEase™ 90W Power Supply features:

  • Constant voltage or current settings
  • Built-in timer for walk-away gel electrophoresis
  • Four sets of output jacks that are compatible with most electrophoresis devices

The PowerEase™ 90W Power Supply accommodates the running of 2 Mini gels, or 2 Midi gels.

The PowerEase™ 90W Power Supply includes the following components:

  • PowerEase™ 90W Power Supply
  • Extra fuse
  • Power cord

Note: Please use the power cord approved for your country/region. 1D Electrophoresis Power Supplies—PowerEase™ 500 Power Supply (discontinued as of December 31, 2013)

The PowerEase™ 500 Power Supply has been discontinued. Our newer alternatives are the PowerEase™ 300W Power Supply and PowerEase™ 90W Power Supply.

Yes, however, keep in mind that the LCD screen will fade away in the cold room. The PowerEase™ would still be working and this will be indicated by the green and red light. The screen should come back on when returned to room temp.

Note: The PowerEase™ 500 Power Supply has been discontinued as of December 31, 2013 and our newer alternatives are the PowerEase™ 300W Power Supply and PowerEase™ 90W Power Supply.

The PowerEase™ 500 Power Supply can be operated under a variety of conditions including constant voltage, constant current and constant power. The default setting is for constant voltage. We recommend running our precast gels at constant voltage for the following reasons:

  • They are continuous gels
  • For most electrophoresis methods, resistance increases throughout the run. When run at constant voltage, current and power decrease throughout the run, providing a natural safety margin and 3) the same voltage setting can be used regardless of the number, or thickness of gels being run.

If you would like to use the PowerEase™ 500 Power Supply to run a gel at constant current, this can be accomplished by setting a voltage limit (at or slightly above the maximum voltage expected to be reached during the run) and then setting the current to the desired level. In this way, the device will try to reach the set voltage. However, when it reaches the threshold current, the current will plateau and the run will switch to constant current (the limiting factor). If no voltage limit is set and a local fault condition occurs, such as a poor connection, very high local resistance may cause the voltage to increase to the maximum of the power supply. This can lead to local overheating and damage to the electrophoresis unit and or gel(s) or create unsafe conditions.

Note: The PowerEase™ 500 Power Supply has been discontinued as of December 31, 2013 and our newer alternatives are the PowerEase™ 300W Power Supply and PowerEase™ 90W Power Supply.

Note: The PowerEase™ 500 Power Supply has been discontinued as of December 31, 2013 and our newer alternatives are the PowerEase™ 300W Power Supply and PowerEase™ 90W Power Supply.

Other than an occasional wipe-down of the instrument to remove salt deposits, which may have built up over time, the PowerEase™ 500 Power Supply does not require a periodic maintenance program.

Note: The PowerEase™ 500 Power Supply has been discontinued as of December 31, 2013 and our newer alternatives are the PowerEase™ 300W Power Supply and PowerEase™ 90W Power Supply.

Using the PowerEase™ 500 Power Supply, the number of Tris-Glycine or NuPAGE™ gels that can be run simultaneously is shown below:

Linear mode is a feature of the PowerEase™ 500 Power Supply that has not been described in the manual. The linear mode can be activated from the 'Edit' screen (where you would change settings for time, voltage, power, current). While on the “Edit” screen, press the left and right arrows simultaneously. This will activate another screen with a three choice menu where the third choice is linear mode. Move the cursor down to 'Linear mode' and turn on Linear mode by using the up/down arrows.

We do offer disposable, empty plastic cassettes and combs to provide the option of pouring your own Mini or Midi gels for use with the XCell SureLock™ Mini-Cell, Mini Gel Tank, or XCell4 SureLock™ Midi-Cell, respectively. All cassettes are sealed on three sides to prevent leaking. Since the cassettes are pre-sealed with the slot pre-taped, a bulky casting stand is not necessary.   • The Empty Mini Gel Cassettes are available in two thicknesses (1.0 mm and 1.5 mm) and can be used with Novex™ Combs that are available in two thicknesses (1.0 mm and 1.5 mm), and in 7 types (1 well, 2 well, 5 well, 10 well, 12 well, 15 well, and 2D well). 
  • The Bolt™ Empty Mini Gel Cassettes are available in 1.0 mm thickness and can be used with Bolt™ 10- or 12-well combs.

We offer the following systems for drying of protein gels:

  • DryEase™ Mini Gel Drying System (Cat. No. NI2387): Simple-to-use kit for even, crack-free drying of mini gels that dries gels evenly, using passive evaporation. Gels dried with this system are suitable for fluorography, densitometry, autoradiography, or permanent gel storage. 
  • Each DryEase™ Mini-Gel Drying System contains the following components:

The pore size of DryEase™ cellophane is 40 microns.

Yes, it is compatible. You need to put the gel in the enhancer solution, wash it and then proceed with the gel drying solution. For detection of signal using 32P, the cellophane is fine. For 35S, the upper layer of cellophane may need to be removed (which is possible) depending on the enhancer being used. For detection of 14C, there is no need to remove the cellophane as it will pass through.

The Gel-Dry™ Drying solution has a proprietary composition.

30% MeOH and 5% glycerol can be tried as a substitute for the Gel-Dry™ Drying solution.

We offer the following staining trays for staining protein gels:

The ZOOM™ IEF Fractionator should be used with an external DC power supply designed for electrophoresis and IEF applications. This power supply must:

  • Be isolated from the ground so that the DC output is floating (safety requirement)
  • Be able to operate at low current (<1 mA) required to perform IEF

Many power supplies automatically shut off when the current drops below 1 mA. You will need a power supply capable of overriding the low current shut-off feature. If the power supply has a current and power limiting capability, it is recommended to set a current limit at 2 mA and a power limit at 2 W.

The electrical leads of the ZOOM™ IEF Fractionator lid are recessed and may not fit into some power supply units. To allow connection of the ZOOM™ IEF Fractionator power leads with certain power supplies, use Novex™ Power Supply Adapters available separately (Cat. No. ZA10001).

Perform fractionation using the following conditions:

100 V for 20 minutes

200 V for 80 minutes

600 V for 80 minutes

The ZOOM™ IEF Fractionator is impervious to alcohol, but not compatible with chlorinated hydrocarbons (e.g., chloroform), aromatic hydrocarbons (e.g., toluene, benzene) or acetone.

We recommend washing the ZOOM™ IEF Fractionator with a mild detergent and rinsing with deionized water after each use. Do not wash lid with cables plugged into the power supply.

We recommend running all applications that require 250 Volts or less on the low voltage/high current side of the ZOOM™ Dual Power Supply.

The ZOOM™ Dual Power supply requires no periodic maintenance program with the exception of an occasional dry wipe-down of the instrument.
