Bioactive proteins such as cytokines and growth factors interact with cells to modulate their behavior. They do this with a certain potency which may be expressed as an EC50, specific activity, or in international units (IU).
An international unit (IU) is a unit of activity for a biological substance that has been tested against an international standard or reference reagent. Potency (IU/ng) of a given biological reference standard is determined by an International Collaborative Study conducted by the World Health Organization (WHO).
Bioactive proteins that have been tested and assigned an internationally accepted unit of biological potency (i.e., IU/ng) are available as international standards or reference materials. Organizations such as the National Institute for Biological Standards and Control (NISBC) test, store, and distribute biological reference standards to allow laboratories across the world to compare their bioactive proteins to these standards.
Bioactive recombinant proteins can have different activities when produced by different labs and protocols, so they should be compared to a reference standard to determine relative potency. The use of IU is intended to simplify comparisons of biological activity of proteins or enzymes obtained from different sources.
Where possible, Thermo Fisher Scientific determines International Unit (IU) values for Gibco PeproTech recombinant proteins through multiple side-by-side comparisons of the PeproTech protein against the analogous WHO international standard or reference standard in a standard biological activity assay. Performing multiple comparison tests helps account for any outliers due to possible variations with the assay. These tests provide a reliable quantification of the PeproTech product’s activity in relation to the activity of the WHO international standard.
The IU values provided are applicable across all lot numbers of a specific catalog number, as testing covers multiple lots of bioactive protein with comparable activity. These values are based on comparison testing and are subject to change.
