Random primers and hexamers are short oligodeoxyribonucleotides with random base sequences (usually [d(N)6]), typically used to prime mRNAs with or without poly(A) for cDNA synthesis. Unlike oligo(dT) primers, these primers are not template specific and are suitable for DNA synthesis using Klenow fragments with DNA templates or for cDNA synthesis using reverse transcriptase with mRNA templates.

Typically, oligo (dT) primers are a string of 12–20 deoxythymidines that specifically anneal to poly(A) tails of eukaryotic mRNAs. In contrast to random primers, oligo (dT) primers are not suitable for degraded samples but are excellent for constructing cDNA libraries and recommended for use with reverse transcriptases (RTs) as Superscript IV RT.
