If you do not designate ROX™ dye as a reference:
- For the 7900 SDS software, the software will not autocall the alleles.
- For the 7500 FAST System SDS software, trailing clusters will appear in the allelic discrimination plots, as shown in the figure below.
Using ROX™ dye as the passive reference dye
7900 SDS Software
Note: The instructions below are for v.2.3 of the software. For other versions of the software, see the user guide.
- Click the well of interest to select it.
- Click the Setup tab.
- At the bottom of the tab, select ROX in the Passive reference list.
- Click green arrow to reanalyze the plate.
7500 FAST System SDS Software
Note: The instructions below are for v.1.3 of the software. For other versions of the software, see the user guide.
- Click the Setup tab.
- In the plate document, double-click the well. The Well Inspector dialog box appears.
- For Passive Reference, select ROX and then click Close.
- Click green arrow to analyze the plate.
ROX™ dye is a passive reference dye that improves the precision of the results by compensating for small fluorescent fluctuations, such as bubbles and small well to well variations that occur in the plate.
