Revolutionary TaqMan® Protein Assays combine the sensitive, specific protein-binding capabilities of antibodies with the superior relative quantitation capabilities of 5' nuclease real-time PCR. They enable fast, easy identification and relative quantification of proteins from small sample sizes or limited numbers of cells-all in a homogeneous assay format. With the TaqMan® Protein Assays Open Kit, you can make your own TaqMan® Protein Assay from biotinylated antibodies for your protein of interest.
Determine protein expression in small samples such as stem cells or germ cell tumors.
Characterize expression of endogenous phosphorylation sites in samples.
Confirm protein expression or characterize induced pluripotent stem cell (iPSC) colonies.
Helpful for validating results from mRNA and/or microRNA (miRNA) assays.
Confirm knockdown of gene targets with siRNAs.
Assays are built from an immunogen-purified polyclonal antibody preparation that is split into two pools or from matched antibody pairs suitable for ELISA, typically made up of one polyclonal antibody and one monoclonal antibody. | Recommended Products |
Lyse cells at a concentration of 1,000 cells/µL cell lysis solution. | Recommended Products |
The assay probes are target-specific antibodies that are conjugated to oligonucleotides through a biotin-streptavidin linkage. Each oligonucleotide in the probe pair presents a 5' or 3' end; these ends are brought into proximity when the antibody components of the assay probe pair bind to two different epitopes on the target protein. | Recommended Products |
The substrate for ligase is a bridge structure formed by hybridization of a third oligonucleotide to the oligonucleotide ends of the assay probe pair. This structure forms preferentially when the assay probes are in proximity to each other. Subsequent protease treatment inactivates the ligase. | Recommended Products |
The ligation product serves as a template for real-time PCR using a TaqMan® Assay for protein detection and relative quantitation. | Recommended Products |
Ct values from the instrument data files are imported into the data analysis oftware for relative quantitation fold change results. | Recommended Products |
