Expression materials and methods: The cDNA ORF of each gene was cloned into pDEST27 expression vector by Gateway cloning, which incorporates a GST tag to the N-terminal of each protein. The proteins were expressed by transfection of the cloned plasmids into Expi293F cell cultures in 96-deep well blocks according to the application note “Expi293 Expression System – Optimizing culture conditions to maximize protein yield and enable use of 96-well microtiter plates.” Expressed proteins were purified from cell lysates using 96-well glutathione-affinity plates and quantitated on a Caliper LabChip 90 (LC90) instrument. Limit of detection of the LC90 assay is 1 mg/L. Protein yields below this limit of detection are shown as <1. Data are presented in order from highest to lowest expression yield.
