When scaling up a T-cell manufacturing process for clinical and commercial manufacturing, it is important to select one that is specifically made for the cell type you are using. Used in >200 clinical trials and in commercialized therapies, the Gibco CTS OpTmizer family of serum-free media (SFM) is thoughtfully designed to meet the nutritional needs of high-density T-cells, which is particularly important when scaling up. The result is that all our media can help maintain the desired central memory phenotype. In addition, whether you’re developing an autologous or allogeneic T-cell therapy, CTS OpTmizer SFM are serum and xeno free and have extensive traceability documentation, including Drug Master Files and Regulatory Support Files.

Material used at every stage of a cell therapy workflow can impact the efficacy of the final drug. Gibco CTS OpTmizer One SFM takes this into account with thoughtfully designed nutrition for T-cells. Not only does CTS OpTmizer One SFM provide the nutrients and energy for T-cell proliferation, it does so without any animal components. This novel animal origin-free formulation helps mitigates the risk of contamination from adventitious agents with superior performance compared to other chemically defined media.
This AOF formulation also demonstrates flexibility and scalability across culture vessels—G‑Rex™ plates, rocking bioreactors, and stir tank bioreactors—CTS OpTmizer One SFM supports scale up while maintaining the desired phenotype. To facilitate scalability, CTS OpTmizer One SFM come in 1 L, 5 L, and 10 L Thermo Scientific Aegis5-14 Film bioprocess containers, which allows compatibility with closed systems.

Gibco CTS OpTmizer T-Cell Expansion SFM, no phenol red is serum-free, xeno-free. Developed for the growth and expansion of human T lymphocytes for autologous therapies. This complete serum-free and xeno-free medium has demonstrated successful T cell expansion in cell culture plates up to large-scale bioreactors as effectively as serum-containing medium.

Gibco CTS OpTmizer Pro SFM is a novel medium, developed for the growth and expansion of human T lymphocytes (e.g., CD4+, CD8+, polyclonal cells, or antigen specific T cells) used in allogeneic cell therapies. CTS OpTmizer Pro SFM improves central memory phenotype and cell growth by shifting the cell’s metabolism. The result is a larger population of central memory cells in a shorter period of time.
CTS OpTmizer Pro SFM come in Thermo Scientific Aegis5-14 Film, which allows compatibility with closed systems. Thermo Scientific Aegis5-14 Film is a five-layer cast film produced in a cGMP facility. The outer layer is a polyester elastomer coextruded with an ethyl vinyl alcohol (EVOH) barrier layer and an ultra-low-density polyethylene product contact layer. Aegis5-14 Film is manufactured using no animal derived components.
