Quality control and minimum yield guarantee

Our quality control processes help increase performance, speed, and capacity. The result is quick and efficient delivery. Below, find additional information about estimated yields based on length of oligo and starting synthesis scale, as well as minimum yields based on synthesize scale and purification option chosen.

When ordering custom oligos, remember that the scale of synthesis (25 nmol, 50 nmol, 200 nmol, 1 µmol, or 10 µmol) is the starting point for synthesis, not the guaranteed final amount. Oligonucleotide length, sequence, GC content, and coupling efficiencies can vary the yield of nmol amounts post-synthesis.

Note: We guarantee the total yield of oligonucleotide as a minimum number of nmol. Purification by HPLC, PAGE, and cartridge will also affect final product yield.

Estimated yields

A universal characteristic of chemical synthesis of oligonucleotides is that the final yield is less than the starting synthesis scale. Despite coupling efficiencies greater than 99%, the iterative addition of each base in the oligo chain compounds the loss of potential full-length material you can achieve. The expected final yield is, therefore, dependent on the length of the oligo. In addition, some material loss also occurs during processing, purification, and quality control. Table 1 shows the estimated nmol yield for desalted oligos at each starting synthesis scale. This table converts the OD units into nmol values for an oligo with an equal and even distribution of each base. The actual nmol conversion from OD for a specific oligo may be different based on the composition and distribution of each base within the oligo.

When ordering custom oligos, remember that the scale of synthesis (25 nmol, 50 nmol, 200 nmol, 1 µmol, or 10 µmol) is the starting point for synthesis, not the guaranteed final amount. The estimated yields above provide guidance on the amount of full-length material we typically achieve, but sequence, GC content, and other factors can vary the actual final yield.
