The TMT11plex Yeast Digest Standard can be used to assess instrument readiness and protein quantification accuracy. The TMT11plex Yeast Digest Standard was used to assess MS instrument readiness on the precision of quantification of a housekeeping protein, ENO2. Protein abundance was scaled to the parental lines in Proteome Discover 2.2 software, with expected ratios of 1:1. A total of 500 ng of the Pierce TMT11plex Yeast Digest Standard was measured using a 50 min gradient on an UltiMate 3000 RSLCnano system with a synchronous precursor selection (SPS)-MS3 (SPS-MS3) method of the Orbitrap Fusion Lumos Tribrid Mass Spectrometer.

Chromatogram of 200 ng Pierce HeLa Protein Digest Standard separated using a Thermo Scientific Acclaim PepMap 100 3 μm x 75 μm x 15 cm column (Product # 160321) with a 2-35% gradient (A: 0.1% FA in water, B: 0.1% FA in 100% acetonitrile) at 300 nL/min for 120 minutes and detected on a Thermo Scientific LTQ Orbitrap XL Mass Spectrometer.
