Achieve in-depth analysis for greater confidence in peak purity and identity with the Thermo Scientific Vanquish Trap Heart-Cut 2D-LC System. This two-dimensional LC (LC-LC) system enables you to collect a single fraction of the first-dimension separation onto a trap column to pre-concentrate, and/or achieve mobile phase compatibility for transferring the fraction for a second-dimension separation or compatibility between UV and MS detection. Heart-cut 2D-LC can reveal hidden impurities that co-elute with the analytes of interest by applying two complementary column chemistries to gain more separation power. The system is configured using modules from the Vanquish Flex UHPLC or Horizon UHPLC systems, delivering performance, robustness and ease of use of the Vanquish LC platform.

Greater confidence

Collect single or multiple fractions of the first-dimension separation onto a trap column and transfer the cut-out fraction(s) to a second-dimension column with complementary column chemistry in an automated workflow for more confidence in peak purity.

Increased sensitivity

The trap column enables enrichment of low-abundant analytes and removal of diluting or disturbing solvents from the eluent by trapping and pre-concentrating the analytes of interest to increase sensitivity.

Simplified method development

The second-dimension eluent provides the flow for solvent modulation, such as eluent strength reduction or buffer exchange of the first-dimension fraction. This process can address solvent incompatibility of UV and MS methods or mobile phase incompatibility between different column chemistries to support multiple combinations of 1D and 2D methods.

The Vanquish Trap Heart-Cut 2D-LC system is fitted with Thermo Scientific Viper fingertight fittings, which support near-zero dead volume connections and enable sharper peaks and tool-free setup.


What is trap-based heart-cut 2D-LC?

Our trap-based heart-cut 2D-LC system allows you to automate and cut out a single fraction eluting from the first dimension (1D) column onto a trap column. This allows for pre-concentrating low-abundant analytes and addresses solvent incompatibility issues before the fraction is eluted onto a second dimension (2D) column to resolve difficult or co-eluting peaks.

 

The flow scheme illustrates how the two six-port valves in combination with the 2D pump and the t-piece are used to reduce the solvent strength and to load the cut-out 1D fraction onto the trap column pre-concentrating the sample before being loaded on the second dimension (2D) column.
