384-well plate high throuhgput gene expression analysis

Gene expression profiles are often used in gene expression-based compound testing because they can indicate pathway activation or inhibition, help monitor complex changes in cellular states, or act as a surrogate for a functional target. QuantiGene Assays are especially useful in secondary compound testing, when more quantitative and robust data are required to provide more detailed information about drug compounds.

QuantiGene assays enable testing groups to:

  • Consistently achieve Z-factors over 0.5
  • Quickly receive your verified custom panel, typically within 3–4 weeks after submitting a gene list
  • Easily automate the simple, ELISA-like workflow in 96- or 384-well plates
  • Precisely detect subtle changes in expression for RNAi knockdown tests

Concentration responses of representative compounds from a confirmation dose-response gene expression-based screen

Figure 1. Concentration responses of representative compounds from a confirmation dose-response gene expression-based test. SW837 cells in this GE-HTS assay were tested at four doses of select compounds ranging from 10 to 0.08 µM for 6 hr (96-well plates; 20,000 cells per well, n = 3 per dose), mRNA levels were measured followed by Log2 fold change calculation per gene and positivity score was determined using the PerkinElmer High Content Profiler. Positivity score was then graphed as a function of compound concentration.
