The BigDye Terminator v3.1 Cycle Sequencing Kit is expertly designed to meet the demands of the vast majority of contemporary applications, including de novo sequencing and resequencing. Consistently generating data with reliably uniform peak heights and optimized signal balance, the BigDye Terminator kit produces long, high-quality reads, and more accurate base assignments for heterozygote and mutation detection.

The chemistry’s powerful formulation is compatible with a variety of templates, including PCR products, plasmids, and large insert clones, such as fosmids and bacterial artificial chromosomes (BACs). However, its dyes are the same as those found in the v3.0 and v1.0/v2.0 kits respectively, meaning no new software or instrument recalibration is required for data analysis. BigDye Terminator v3.1 and v1.1 cycle sequencing kits are completely comprehensive and include all required reagents for sequencing 24, 100, 1,000, 5,000, or 25,000 single stranded (ss) or double-stranded (ds) DNA templates. The reagents in each kit are optimized for use with the 3730xl, 3500, SeqStudio, and Seqstudio Flex genetic analyzers.

Application Note: A Workflow for Obtaining High Quality Sequencing Data from Bacterial Artificial Chromosome (BAC) DNA

Bacterial artificial chromosome (BAC) DNA is difficult to use as a sequencing template, because of its large size (usually >100,000 bp). A modified sequencing protocol has been demonstrated for BAC DNA using the BigDye Terminator Cycle Sequencing Kit. The BigDye Terminator v1.1 and v3.1 chemistries can be diluted, and a 5X Sequencing Buffer is provided with the kit to enable you to do so. 

The BigDye Direct Cycle Sequencing Kit and the dGTP BigDye Terminator Ready Reaction Kits v1.0 and v3.0 cannot be diluted.

Possible causes could be oxidation or a change in pH when injecting out of water or old Hi-Di formamide that has broken down. Some cleanup methods can also oxidize the dyes. Alternatively, an old or contaminated array can cause dye breakdown, while an arcing event on the system can destabilize the blue baseline and, in severe cases, mask it completely.

It’s all about choosing the right chemistry for your application. If your research requires the most robust, flexible chemistry that will generate the longest reads, then the v3.1 kit is most suitable. If your laboratory is primarily sequencing short PCR fragments using rapid electrophoresis-run modules, then the v1.1 kit would serve you better. Many variables contribute to DNA sequencing data quality, including template type, instrument module, total signal, peak-height uniformity, and mobility shift. All should be taken into consideration when selecting the most appropriate chemistry for your work.
