Mach1 T1R is the fastest-growing chemically competent strain currently available (Figure 1). Their colonies are clearly visible within eight hours of plating the transformed cells (ampicillin selection only). Mach1 T1R competent cells are phage resistant owing to the tonA (fhuA) deletion in their genotype and are designated as Mach1 T1R. Phage resistance properties protect cells from lysis by T1, T5, and φ80 phages.
Figure 1. The rapid growth rate of Mach1 T1RE. coli compared to standard cloning. Mach1 T1R, XL10-Gold, TOP10, XL1-Blue, and DH5α E. coli were transformed with pUC19 and grown overnight. Each culture was inoculated 1:50 in 50 mL of LB ampicillin in 500 mL flask. Growth was measured every 30 minutes for 5.5 hours. Mach1 T1R cells reached a desired optical density (e.g., for mini-prep analysis) significantly faster (in 4 hours) than other tested strains.
